Nitrogen-fixing organelle in a marine alga

Studying a marine alga with a cyanobacterial endosymbiont, Coale et al. used soft x-ray tomography to visualize cell morphology and division of the alga, revealing a coordinated cell cycle in which the endosymbiont divides and is split evenly, similar to the situation for plastids and mitochondria in these cells.


Tree water uptake patterns across the globe

knowledge of how the distribution of water uptake depth (WUD) varies across species, climates, and seasons is scarce relative to our knowledge of aboveground plant functions. With a global literature review, Bachofen et al. found that average WUD varied more among biomes than plant functional types (i.e. deciduous/evergreen broadleaves and conifers), illustrating the importance of the hydroclimate, especially precipitation seasonality, on WUD.


Imaging of labile Fe2+ and Fe3+ in living Arabidopsis thaliana roots

Xiong et al. selected two fluorescent probes, SiRhoNox-1 and MPNBD, which they used to image labile Fe2+ and Fe3+, respectively, in Arabidopsis (Arabidopsis thaliana) roots.

Zygnematophyceae are the algal sisters of land plants. Feng et al. sequenced four genomes of filamentous Zygnematophyceae, including chromosome-scale assemblies for three strains of Zygnema circumcarinatum.

Plasmodesmata provide routes for the movement of various molecules between plant cells. Cargo molecules of plasmodesmata include metabolites like sucrose, small signaling molecules like hormones, and large endogenous signaling molecules like mRNAs.

The temperature sensor TWA1 is required for thermotolerance in Arabidopsis 

Bohn et al. show that TWA1 is a temperature-sensing transcriptional co-regulator that is needed for basal and acquired thermotolerance in Arabidopsis thaliana. At elevated temperatures, TWA1 changes its conformation and allows physical interaction with JASMONATE-ASSOCIATED MYC-LIKE (JAM) transcription factors and TOPLESS (TPL) and TOPLESS-RELATED (TPR) proteins for repressor complex assembly.


Widespread horizontal gene transfer between plants and bacteria

Haimlich et al hypothesized that the spatial vicinity and the long-term relationships between plants and their microbiota may promote cross-kingdom horizontal gene transfer (HGT), a phenomenon that is relatively rare in nature. To test this hypothesis they analyzed the Arabidopsis thaliana genome and its extensively sequenced microbiome to detect events of horizontal transfer of full length genes that transferred between plants and bacteria.


Plasmodesmal closure elicits stress responses

Plant cells are connected to their neighbors via plasmodesmata facilitating the exchange of nutrients, hormones, and signaling molecules. While immune elicitors trigger changes in plasmodesmal aperture, it is unknown how this contributes towards a full immune response. To investigate this, Tee et al. developed two transgenic lines with which we could induce plasmodesmal closure independently of immune elicitors.

CRISPRi-based circuits to control gene expression in plants

Khan et al create a toolkit of engineered repressible promoters of different strengths and construct NOT and NOR gates in Arabidopsis thaliana protoplasts. They determine the optimal processing system to express single guide RNAs from RNA Pol II promoters to introduce NOR gate programmability for interfacing with host regulatory sequences. The performance of a NOR gate in stably transformed Arabidopsis plants demonstrates the system’s programmability and reversibility in a complex multicellular organism.
